RESUMO
Chromobacterium violaceum é um bacilo de vida-livre, Gram-negativo
comumente encontrado no solo e nas águas de regiões tropicais e subtropicais.
Uma das principais características deste organismo é sua capacidade de
produzir o pigmento violaceína, o qual apresenta inúmeras atividades
biológicas. Em 2003, o genoma deste organismo foi completamente
sequenciado e revelou informações importantes sobre a fisiologia desta
bactéria. Porém, poucos estudos pós-genômicos tem sido realizados. Este
trabalho avaliou o perfil proteico de C. violaceum cultivada em meio LB a 28ºC,
o que permitiu a identificação de proteínas relacionadas a um possível sistema
de secreção ainda não identificado e caracterizado em C. violaceum, ao
sistema quorum sensing, a processos regulatórios da transcrição e tradução,
adaptação ao estresse e ao potencial biotecnológico. Além disso, a resposta
desta bactéria à radiação UVC foi avaliada. A comparação do perfil protéico,
analisado por eletroforese 2-D, do controle versus tratado possibilitou a
identificação de 52 proteínas que surgiram após a indução do estresse. Os
resultados obtidos permitiram a elaboração de uma via de resposta de C.
violaceum ao estresse gerado pela luz UVC. Esta via, que parece ser de
resposta geral ao estresse, envolve a expressão de proteínas relacionada à
divisão celular, metabolismo de purinas e pirimidinas, choque térmico ou
chaperonas, fornecimento de energia, regulação da formação de biofilme,
transporte, regulação do ciclo lítico de bacteriófagos, além de proteínas que
ainda não apresentam função caracterizada. Apesar da reposta apresentar
similaridades com a SOS clássica de E. coli, ainda não podemos afirmar que C.
violaceum apresenta uma resposta SOS-like, principalmente devido a ausência
da caracterização de um proteína LexA-like neste organismo.

ABSTRACT
Chromobacterium violaceum is a free-living bacillus, Gram-negative commonly
found in water and sand of tropical and subtropical regions. One of its main
characteristic it's the ability to produce the purple pigment named violacein, that
shows countless biological activities. In 2003, the genome of this organism was
totally sequenced and revealed important informations about the physiology of
this bacteria. However, few post-genomics studies had been accomplished.
This work evaluated the protein profile of C. violaceum cultivated in LB medium
at 28ºC that allowed the identification and characterization of proteins related to
a possible secretion system that wasn't identified and characterized yet in C.
violaceum, to the quorum sensing system, to regulatory process of transcription
and translation, stress adaptation and biotechnological potential. Moreover, the
response of the bacteria to UVC radiation was evaluated. The comparison of
the protein profile, analyzed through 2-D electrophoresis, of the control group
versus the treatment group allowed the identification of 52 proteins that arose
after stress induction. The obtained results enable the elaboration of a stress
response pathway in C. violaceum generated by the UVC light. This pathway,
that seems to be a general stress response, involves the expression of proteins
related to cellular division, purine and pirimidine metabolism, heat chock or
chaperones, energy supply, regulation of biofilm formation, transport, regulation
of lytic cycle of bacteriophages, besides proteins that show undefined function.
Despite the response present similarities with the classic SOS response of E.
coli, we still cannot assert that C. violaceum shows a SOS-like response, mainly
due to the absence of characterization of a LexA-like protein in this organism.

6. CONSIDERAÇÕES GERAIS
Os resultados obtidos nos dois manuscritos apresentados auxiliaram no
entendimento mais detalhado acerca do metabolismo basal de C. violaceum e
nas mudanças deste metabolismo frente ao estresse gerado pela radiação UV.
Os artigos são pioneiros no estudo proteômico deste organismo e abre
possibilidades no que se refere aos estudos pós-genômicos realizados até
então.

7. PERSPECTIVAS
Outros experimentos além dos apresentados nesta tese foram
realizados. Entre eles podemos citar a proteômica de uma cepa mutante no
operon de síntese da violaceína, exposta ou não a radiação UVC.
A proteômica desta cepa mutante sem exposição à radiação UVC foi
realizada com o intuito de auxiliar no entendimento da função deste pigmento
no contexto fisiológico bacteriano. O gel de referência das proteínas desta
bactéria mutante apresentou 365 spots. Quando comparado com o gel de
referência da cepa selvagem (Artigo: Medeiros, et. al., 2011) foram observados
apenas 165 spots em comum. Os resultados da expressão diferencial na
ausência da violaceína mostram que este pigmento bloqueia a produção de
algumas proteínas, todavia na sua ausência surgem 200 spots, os quais já
foram tripisinisados e estão sendo identificados por MS.
A exposição da cepa mutante à radiação UVC também foi realizada. No
controle foram identificados 148 spots e 243 no tratado, onde apenas 81 spots
foram comuns a ambas. Todos os spots que sugiram após o tratamento foram
tripsinisados e 73 foram identificados, correspondendo a 57 proteínas
diferentes. A análise da resposta da bactéria na ausência da violaceína poderá
ser útil para verificar a existência de um possível papel protetor deste pigmento
frente ao estresse gerado pela luz UVC.
Estes resultados possibilitarão a publicação de, pelo menos, mais dois
artigos científicos oriundos desta tese de doutorado.
