SCREENING MODELS
in vitro and ex vivo models
in vitro enzymatic cell-free pre-screening performed with the rate-limiting enzyme of the
target process in the skin (i.e. tyrosinase in melanin production)
in vitro on cell lines advantage of immortalised cell lines: proliferate indefinitely
in vitro on primary human cells cells cultured directly from a subject; lifespan is limited, but closer to
human skin
in vitro on reconstructed human skin model (3D model) primary human cells cultured on an inert medium forming a cornified
epidermal layer;
consists of keratinocytes and optionally other cells (melanocytes,
fibroblasts, dendritic cells etc.)
ex vivo on excised human / pig skin model skin pieces are excised out of surgical skin
advantage over 3D model is the presence of all skin types in the native
structural organisation, including hair follicles and adipose tissue
