Cerebralcare Granule (养血清脑颗粒) , one of the famous classical recipes in

traditional Chinese medicin, is developed from the "Decoction of Four Drugs". It is able

to ameliorate the detrimental effect on cognition following chronic cerebral

hypoperfusion , improve cerebromeningeal microcirculation, inhibit platelet

aggregation, and release vasospasm. Currently, the recipe is mostly used for treating

headache, including migraine headache, neurogenic headache, etc.

Hypertension (高血压) is a cardiovascular disease with the most

epidemiological impact in the world and also represents a major risk factor for

developing other diseases as endothelial dysfunction, metabolic syndrome,

diabetes, renal dysfunction, congestive heart failure, coronary artery disease

and stroke.

Worldwide hypertension is estimated to cause 7.1 million premature deaths

and 4.5 % of the disease burden.

通过超高效液相色谱建立了快速、高效分离分析 4 个酚酸的方法,绿原酸 396.17±42.05 ,咖啡酸 81.67±9.55 ,阿魏酸 251.17±28.25 ,迷迭香酸 582.41±88.14 µg/g 。

用气相色谱( GC-MS )法建立指纹图谱,对其中化学成分进行分析鉴定。 10 批养血清脑颗粒得到的共有指纹图谱选定了 10 个共有峰,通过质谱鉴定出 10 个共有峰的化学成分。

5. 脑循环改善

养血清脑颗粒抑制 右旋糖酐引起小鼠大脑皮质血流量减少,改善脑组织血流量,改善脑组织微循环,缓解脑组织供血不足所致的各种病证。明显改善慢性脑缺血大鼠局部脑血流量,提高学习记忆能力。

6. 脑神经保护

养血清脑颗粒对两侧颈总动脉结扎后再灌注损伤模型,能选择性抑制谷氨酸、 caspase-3 的表达,并且通过谷氨酸盐合成酶选择性抑制兴奋性氨基酸谷氨酸的神经毒性作用,减少神经细胞的死亡,从而发挥神经保护作用。

体外吸收:比较了阿魏酸和芍药苷的体外吸收特性;体内吸收:对全方给药后延胡索乙素药代动力学进行了研究;代谢:未对肝微粒体酶及对同服其它药物体内代谢过程产生影响。

养血清脑颗粒能浓度依赖性地拮抗由 NE 引起内皮完整和去内皮的血管收缩,对内皮完整血管的舒张作用强于去内皮血管。养血清脑颗粒舒张血管具有内皮依赖性非内皮依赖性和浓度依赖性。

分别用内皮细胞舒张因子 eNOS 抑制剂 L-NAME 和前列环素途径中环氧合酶抑制剂 INDO孵育胸主动脉环,发现二者均能使养血清脑颗粒的舒血管作用减弱。

养血清脑颗粒对 Kir 和 KCa 通道无影响,能使得 KATP 通道开放。

不同浓度的养血清脑颗粒使得 CaCl2 量效曲线右移,推测其对血管的舒张作用主要是通过外钙内流而发挥作用的;预先用养血清脑颗粒孵育血管环后,再用 NE刺激血管,血管收缩作用减弱,说明其舒血管作用与肌浆网储存钙释放有关
