“Millions of our cells die through apoptosis every day”

Gabriel Gil leads the Apoptotic Sig-nalling research group of the IMIM since 2000. Apoptosis, or program-

med cell death, is a physiological process with an essential role both during deve-lopment and in the adult organism. “The epithelium of our body renews each day, in such a way that millions of cells die through apoptosis daily. Self-reactive lymphocytes, which recognise the body’s own antigens and which can attack it, are also eliminated by apoptosis”, explains the researcher. The absence of apoptosis, on the other hand, is a trait characteristic of the majority of tu-mours. “Apoptosis is, in fact, a safeguard mechanism existing in all cells and which ensures that if the DNA becomes damaged and impossible or very difficult to repair, the cell will die, meaning this genetic error will not be passed on. If this mechanism does not work, the errors are perpetua ted, as happens in cancer”, says Gil.

A new cyclin

Soon after he set up his group, Gil identified a new protein, cyclin O, which appears to be implicated in the detection of DNA lesions. They believe that it could be the link between the detection of these lesions and the initia-tion of the apoptotic mechanism. Since then, this protein has been the focus of their rese-arch and they have discovered that it is ove-rexpressed in various cancers, especia lly of the colon and bladder. “We work with patho-

logists, oncologists, dermatologists, and uro-logists from the Hospital del Mar in order to study, in patient samples, why cyclin O is not regulated in these cases and what advantages this offers the tumour”, says Gil. To understand how cyclin O works on a molecular level, the group uses diffe-rent biochemical and molecular biology techniques. They also work with animal models and have generated the first mou-se knockout (KO) for cyclin O, a mouse which does not express this protein. Cyclin O is expressed only when the cell becomes damaged. Over the past years the group has discovered some of the external stimuli which provoke apoptosis via cyclin O, such as DNA damage or endoplasmic re-ticulum stress through the accumulation of badly folded proteins.

Alongside the patients

Recently the group has discovered that cyclin O can regulate ATM, a protein es-sential for the detection of DNA damage. ATM mutations are the cause of ataxia te-langiectasia, a very rare but very serious disease which causes immunodeficiency, neurological problems and a greater risk of developing tumours. Gil received a grant from La Marató de TV3 to study the role of cyclin O in this disease. Recently, the Spanish associati-on for families with ataxia telangiectasia (AEFAT) invited the scientist together with Óscar Fernández-Capetillo, a researcher from the CNIO and an expert in the field of ATM, to present their research to the fami-lies of those affected. The researchers we-re then asked to help in preparing a simple and standardised molecular diagnostic method for the disease. “The problem is that there is no established molecular di-agnostic procedure, and the complexity of the gene and the low frequency of the disease make it commercially unattrac-tive to develop one. But being able to di-agnose it within the first year of life may greatly improve the results of the palliative treatments which these children currently recei ve”, emphasises the head of the group. “In addition, it would be very useful to identify the carriers of the mutated gene, who, even though they do not suffer the disease, also have a higher probability of developing tumours”, he says ■

Alzheimer’s, RNA viruses and oxidative stress

Astudy published in the journal PLoS ONE and coordinated by Francisco Muñoz (CEXS-UPF) re-

veals new data about the activation of the BACE1 enzyme. This enzyme is responsi-ble for the production of the beta-amyloid peptide, the aggregation of which leads to Alzheimer’s. The researchers show that BACE1 is activated in the neurons due to the action

of the PKR enzyme through factors which generate cell stress, such as the presence of double-stranded RNA viruses or oxidative stress. “Human ageing generates free ra-dicals which damage proteins, lipids, car-bohydrates and nucleic acids (DNA and RNA). This damage, together with double-stranded viruses such as the herpes sim-plex virus HSV1, are present in the brains of the majority of old people and contribu-te to PKR activation”, explains Muñoz. PKR phosphorylates the initiation factor eIF2al-fa, which triggers the synthesis of various proteins, including BACE1. “There are four enzymes which activa-te BACE1 in the brain, and PKR is the only one that acts pathologically in response to cell stress. Understanding the biochemical basis of this type of activation should help in the development of preventative medi-cines”, states Muñoz ■

Common origin for genetic diseases

An analysis of molecular networks has revealed that many diseases, including Mendelian ones, also

known as “monogenic”, cannot be attri-buted to alterations of a single gene but rather are associated with changes in va-rious genes. The study also shows that the diseases have a great deal of interconnec-tion, and suggests a common genetic ori-gin of Mendelian, complex and environ-mental diseases. The work was carried out by a team of researchers from the GRIB (UPF-IMIM), in collaboration with the University of Mu-nich in Germany, and has been published in the journal PLoS ONE. The network analysis was undertaken using the DisGeNET software, a tool deve-loped by Laura Furlong's team at the GRIB. The databases analysed span the entire spectrum of human diseases with a genetic

component: Mendelian or monogenic di-seases, complex diseases caused by many genes, and those diseases having also envi-ronmental causes. "We made a systematic study of gene-di-sease networks to see if the genes involved

were related functionally in the cell, that is, if they formed part of molecular networks or functional modules. This is, in fact, true, and moreover they are genes that are usua lly part of a set of basic biological pro-cesses", explained Furlong ■

pluripotent stem cells (iPS), although the mechanisms are neither very efficient nor well understood. Researchers at the CRG, led by Pia Cosma, have taken an important new step into the dissection of the somatic cell reprogramming mecha-nism. The authors had previously demonstra-ted that activation of the Wnt/β-catenin signalling pathway triggers somatic cell reprogramming. Now they have shown that the absence of T-cell factor 3 (Tcf3), a repressor of β-catenin target genes, stri-

kingly and rapidly enhances the efficiency of neural precursor cell (NPC) reprogram-ming. The study, published in PNAS, also shows that the increased reprogramming efficiency is largely due to genome-wide epigenetic modifications. “The possibility of obtaining pluripo-tent stem cells from adult somatic cells, although still far from being a therapeutic reality, promises to overcome fundamen-tal issues in stem cell therapy, including ethical concerns about the use of human embryonic stem cells”, says Cosma. “Bet-ter knowledge of the mechanisms that regulate reprogramming will, without doubt, help to develop safer procedures to obtain iPS for clinical use”, she adds ■

CONFERÈNCIES PRBB-CRG / CONFERENCES PRBB-CRG Conference Programme financed by the CRG and the PRBB

RUI COSTA, Friday September 9. Costa, from the Instituto Gulbenkian de Ciência in Oeiras, Portugal is studying the neurobiology of action, particularly that which is learned through trial and error through-out life, in health and disease. In particular, he tries

to understand how changes in molecular networks in the brain modify neural circuits to produce experience-dependent changes in actions. In order to understand this he focuses on three dif-ferent components: action initiation, how we start and generate diverse actions; action improvement, how we increase the ac-curacy and speed of actions through trial and error; and actions and outcomes, how we learn that particular actions lead to par-ticular outcomes (goal of the action) and how we form habits. He has been invited by Filipe Pinto Teixeira (CRG).

KENNETY M. WEISS, Monday September 19. Weiss, from the Evolutionary and Developmental Genetics group of the Department of Anthropology at the Penn State University, USA, uses a variety of investigative techniques for understanding complex

traits and the evolutionary processes that are responsible for them. He works on various subjects, ranging from understan-ding the amount of genetic complexity underlying human di-sease, to developmental and evolutionary aspects of the head and dentition. He is also interested in understanding complex traits that seem to be affected by multiple factors and in using genomic sequence comparisons to understand gene regulation and evolution of the olfactory system, amongst others. He has been invited by Jaume Bertranpetit (UPF).

FREDA MILLER, Friday September 16. Miller, from the Developmental & Stem Cell Biology depart-ment of the Hospital for Sick Children Research Institute and the University of Toronto in Canada, is interested in the role stem cells play in the bio logy

of adult tissues and in using adult stem cells to study and treat diseases. Known for her studies of neuronal stem cells and of neuronal growth, survival and apoptosis, other major findings from her lab have provided evidence that mammalian dermis contains a multipotent stem cell population (SKPs) that can be isolated and purified, that the p75 neurotrophin receptor is apoptotic in neurons, and that the p53 family plays an essential role in the nervous system. She has been invited by Bill Keyes (CRG).

Cristina Gómez Santos, born in Barcelona 33 years ago, splits her time between caring for her children (a two-and-a half year old baby girl and a five-month old baby boy) and her job looking after the CMRB researchers. After brief stays in academia and industry she became lab manager of the CMRB labs in 2007.

Trobes a faltar fer recerca?

No tenia clar què volia fer, i tenir dues feines era molt cansat.

CARRERA CIENTIFICA / BECOMING A SCIENTIST

CRISTINA GÓMEZ SANTOS – RESPONSABLE DELS LABORATORIS DEL CMRB

“My work is different everyday and I help out the researchers”

When did you leave research?

After doing my PhD on the neuros-cience of Parkinson’s, I started tea-ching lab technicians in the Escola

Bonanova in the mornings and working as a cytogeneticist in the Laboratorios Dr. Eche-varne in the afternoons. There I was introdu-ced to the world of genetic diseases.

What were these experiences like?I loved teaching, but in private laborato-ries everything is very routine. After a ye-ar I changed to the Balagué Center labs, which were bigger and did more things.

But you left after six months.I've always liked doing new things. I wasn’t sure what I wanted to do and having two jobs was really tiring. When the opportunity came up for me to be a product specialist at the multinational Olympus, I went for it.

What did your job consist of?Promoting the microscope software for

doing cytogenetics and in vitro fertilisation. I had to travel a lot in Spain and abroad to give courses and seminars. I learnt a lot and I got to know all of the genetics and assisted reproduction labs in Spain.

How did you arrive at the CMRB? I already used to come here with my job, they knew me as “the Olympus girl”, and I really liked this centre and all of the PRBB. Looking on the CMRB webpage I saw the job offer and thought, why not?

What are your responsibilities? To coordinate the CMRB laboratories and equipment rooms. I supervise the work of the technicians, control the stock of ma-terial and general reagents, ensure that all of the equipment is in working order, deal with sales reps, test the new kits and look af-ter the researchers for whatever they need.

It seems that the CMRB couldn’t function without you...Nor without the technicians either! More-over, apart from me there are the heads of the different technical platforms: Bioima-ging and Histology, Cell Culture, Flow Cyto-metry, and Embryo Micromanipulation.

Do you miss doing research?I loved doing my thesis, but research is very competitive and stressful, with no holidays and without timetables. Here I am lucky that, apart from managing, I can also do lab work side-by-side with the technicians and I am up to date with the research that is do-ne at the CMRB thanks to our monthly lab meetings.

What do you like most about your job?Every day is different, very full, and I help the researchers. I am in contact with the lab, which is the thing I like most, and I meet people from many countries, it is very en-riching, Moreover, since I had my children, I work an intensive schedule, so the timeta-ble is also perfect. And I live really close by!

What does being within the PRBB offer?Due to the huge number of researchers at the PRBB, lots of sales reps come to show us the latest technology and equipment which is perfect! I think that I am very lucky to be here, and I hope to be able to stay here for a long time! ■

A gene for Bohring-Opitz syndrome identified

A cause of Bohring-Opitz syndrome, a very serious disease characterised by severe intellectual retardation and

The study used massive sequencing techniques to simultaneously look for muta-tions in 20,000 genes. This search identified,

Fifteen years later, the rese-archers have found that anxiety, including social phobia and agoraphobia, has pri-marily manifested in those with hypermo-bility. Hypermobility is a benign disorder of the musculoskeletal system, associated with an increase in flexibility of the body’s connecting tissue due to an alteration of the collagen fibre.

Quin talent t’agradaria tenir?M'encantaria saber explicar acudits; se'm dóna fatal!.

What are your hobbies?I really like, in this order: playing volleyball, music, reading and architecture.

What would make you absolutely happy?Everyone being able to appreciate how beautiful it is to see the sun rise every morning.

What bothers you most?Arrogance! In the end we all have good and bad traits, and above all, things that make us special! Nobody is better than anybody else.

Who is your favourite real or fictitious person?Actually, I really admire my mum: I think she’s truly extraordinary.

What is your worst failing?I am unable to remember anyone’s name, even if I am told a thousand times.

What talent would you like to have?I would love to be able to tell jokes. I am quite bad at it!
