Reactions 1459 - 6 Jul 2013 S Pregabalin Various toxicities: 3 case reports Three women developed various toxicities while receiving pregabalin [routes not stated] to treat restless legs syndrome (RLS), fibromyalgia syndrome and neuropathic pain. A 42-year-old woman was diagnosed with RLS and fibromyalgia syndrome, and started pregabalin 75mg twice daily. After 2 days, she presented with complaints of sedation and vertigo. The woman was advised that these side effects would resolve with time. At follow-up 15 days later, she reported that the sedation and vertigo had resolved, and her RLS symptoms had improved by > 50%. Her pregabalin dose was subsequently increased to 150mg twice daily, with > 90% relief of her RLS and pain symptoms. A 38-year-old woman was diagnosed with RLS and diabetic neuropathic pain, and subsequently commenced on pregabalin 75mg twice daily; she also had lumbar disc herniation. She was admitted 1 week later, due to complaints of vertigo, fatigue and a tendency to sleep. These side effects resolved within 1 week. Her pregabalin dose was subsequently increased to 150mg twice daily, improving her RLS symptoms by 80%. A 48-year-old woman was diagnosed with RLS and myofascial pain syndrome, and was started on pregabalin 75mg twice daily [duration of therapy before reaction onset not clearly stated]. She was advised about side effects to pregabalin, such as dizziness, sleepiness and vertigo. At her follow-up visit 1 week later, the woman reported experiencing similar side effects initially, but said they had resolved within 4–5 days. Her pregabalin dosage was subsequently increased to 150mg twice daily, complete relief of her RLS and pain symptoms after 3 months. Author comment: "Side effects, such as headache, vertigo, rash, dry mouth and a tendency to sleep may be observed, particularly with initial doses of pregabalin, but resolve with time.
