What is a spot test?
A spot test is …a simple chemical reaction that allows a substance to be identified.

In drug testing…• a drug or class of drugs gives a characteristic visible change
when mixed with another chemical (the reagent)• a sample, such as urine, is tested with the reagent• the presence of the drug (or class of drug) is evidenced by the
characteristic visible change; this is a positive test• if the characteristic change is not seen, it is a negative test

1. Place the culture plate on a white piece of paper2. Mark the columns and rows, as in the diagram3. Place a drop of urine sample A into the top two wells in column A4. Place a drop of urine sample B into the top two wells in column B5. Place a drop of urine sample C into the top two wells in column C6. Add drops of reagent Y to row Y, and drops of reagent Z to row Z
What you have to do
Work in pairs. One of you should use the tophalf of the well plate. The other should repeatthe procedure in the bottom half.

The colour identifies the class of drug. The intensity of the colour is an approximate measure of how much of the drug is present.

• don’t tell you how much of the banned substance is present• don’t give a permanent record of the test result

Modern instruments can identify the actual drug present, an accurate measure of how much of it is present, and they provide a permanent record.
