1. Úvod
Cytoskelet je vysoce dynamická trojrozměrná síť proteinů, která je základní složkou všech
eukaryotických buněk. Cytoskeletální vlákna, tj. mikrofilamenta a mikrotubuly, zajišťují přenos
signálů, tok informací i stavebního materiálu, pohyb nebo kotvení proteinů a organel v buňce, ale i
pohyb celých buněk a organismů. Proteinové polymery cytoskeletu protínají cytoplasmu buněk,
kde plní své četné funkce, uplatňují se při dělení buněk, růstu a vývoji pletiv a orgánů. Funkční
cytoskeletální síť musí podléhat neustálé reorganizaci. Řízená výstavba, odbourávání a transport
podjednotek cytoskeletálních vláken se účastní všech životních projevů buněk. Reorganizace
cytoskeletu se účastní i tzv. asociované proteiny, které jsou nezbytné pro jeho správnou funkci.
Cytoskelet je struktura velmi konzervovaná, většina poznatků platí tedy pro živočišné i rostlinné
buňky, existují však i funkce a struktury typické pouze pro rostliny.
Cytoskelet se účastní veškeré morfogeneze rostlin (pro přehled viz Mathur a Hülskamp,
2002). Rostlinné buňky jsou nepohyblivé a jejich uspořádání v pletivech a tvar rostlinných orgánů
jsou dány přesně orientovaným dělením a řízeným růstem buněk. Na obou těchto procesech se
podílí jak mikrofilamenta, tak mikrotubuly. Cytoskelet současně zajišťuje také buněčnou
morfogenezi, tvorbu složitých tvarů jednotlivých, často různě specializovaných buněk (Mathur,
2006). Během všech růstových a vývojových procesů je zároveň nezbytná vzájemná interakce a
koordinace obou zúčastněných cytoskeletálních struktur. Té se účastní některé proteiny
asociované s cytoskeletem (pro přehled viz Petrášek a Schwarzerová, 2009). Protože jsou rostliny
jako celek organizmy vázané na jedno stanoviště, jsou na flexibilitu cytoskeletu kladeny vysoké
nároky také z hlediska reakce na podněty z prostředí.
Díky své významné úloze v buňkách cytoskelet je velmi intenzivně studován. Rozvoj
poznatků umožnil zejména nástup moderních biochemických, mikroskopických a zvláště
molekulárně biologických metod. Ale právě kvůli obrovské šíři dějů, kterých se cytoskelet
účastní, zůstává stále řada neznámých. V této práci jsem se pokusila přispět k objasnění úlohy
cytoskeletu a proteinů s ním spolupracujících v několika situacích dotýkajících se morfogeneze
rostlinných buněk i celistvých rostlin. Práce je členěna do čtyř nezávislých kapitol, jejichž
pojítkem je úloha cytoskeletu v různých morfogenních procesech, jimiž jsou interakce
mikrofilament a mikrotubulů, působení růstového retardantu ancymidolu v kontextu tvorby
buněčné stěny, role aktinu ve vývoji somatických embryí a její ovlivnění latrunkulinem B a reakce
buněk kořenů na toxické působení hliníku.

2. Úloha ARP2/3 komplexu v interakci mikrofilament a mikrotubulů
Mikrofilamenta a mikrotubuly tvoří v buňce dvě nezávislé a přesto velice úzce provázané
sítě, jejichž vzájemná interakce je oblastí s řadou neznámých. ARP2/3 je konzervovaný
proteinový komplex, složený se sedmi podjednotek, asociovaný s mikrofilamenty (Goley a Welch,
2006; Mathur, 2005). Jeho primární funkce je spojena s dynamikou aktinové sítě (nukleace,
větvení vláken aktinu), ale zdá se, že ARP2/3 komplex by mohl být zapojen i do interakce mezi
mikrofilamenty a mikrotubuly. Ke studiu funkce a lokalizace tohoto komplexu u rostlin jsem
využila jednu z jeho podjednotek, protein ARPC2, který by mohl být pojítkem mezi
mikrofilamenty a mikrotubuly, a tabákovou buněčnou linii BY-2.
Z buněk BY-2 byla získána sekvence NtARPC2, která byla dále využita k izolaci proteinu,
přípravě protilátky a fúzována s GFP.
Fúzní protein GFP-APRC2 tranzientně overexprimovaný v buňkách BY-2 vytvářel síť
vláken, která částečně kolokalizovala s mikrotubuly, méně však s aktinem.
Kosedimentační analýza proteinu ARPC2 s aktinem a mikrotubuly in vitro ukázala, že
protein ARPC2 je schopen přímé vazby jak na aktinová filamenta, tak na mikrotubuly.
Vizualizace ARPC2 a ARP2 podjednotek ARP2/3 komplexu v buňkách BY-2 pomocí
nepřímé imunofluorescence ukázala jejich lokalizaci ve formě drobných, charakteristicky
uspořádaných teček, které dekorovaly mikrofilamenta a kolokalizovaly i s mikrotubuly.
Uvedené výsledky naznačují, že ARP2/3 komplex má kromě své nukleační aktivity ještě
další funkce. Mohl by sloužit jako kroslinkující faktor mezi aktinem a mikrotubuly. Za vazbu na
mikrotubuly by pak mohla být přímo zodpovědná ARPC2 podjednotka komplexu.
3. Ancymidol jako inhibitor biosyntézy celulózy
Ancymidol je retardant rostlinného růstu s inhibičním účinkem na biosyntézu giberelinů,
jehož hlavním efektem je redukce dlouživého růstu rostlin. Byla však popsána také tvorba
tvarových malformací ke giberelinům necitlivých buněk BY-2 vlivem působení ancymidolu
(Boříková et al., 2003). Protože tvar udává rostlinným buňkám buněčná stěna, zaměřili jsme při
studiu působení ancymidolu právě na ni. Majoritní složka buněčné stěny, celulóza, je
syntetizována vně buněk celulóza-syntázovými komplexy (Cosgrove, 2005). Jejich pohyb a s ním
související směr ukládání celulózních mikrofibril buněčných stěn je řízen mikrotubulárním
cytoskeletem (Wasteneys a Fujita, 2006). Ke studiu na giberelinech nezávislého mechanizmu
účinku ancymidolu byly využity buňky BY-2 a semenáčky tabáku Nicotiana benthamiana.
Ancymidol způsoboval v koncentracích 10 -100 µM tvarové změny buněk BY-2. Míra
těchto změn byla úměrná použité koncentraci, s přibývajícím počtem a velikostí malformací
klesala viabilita buněk. V 1 mM koncentraci ancymidolu však nedocházelo k tvorbě výdutí, ale k

inhibici prodlužovacího růstu buněk a poruchám tvorby nové buněčné přepážky po rozdělení
buněk. Tyto efekty ancymidolu nesouvisely s jeho antigiberelinovým působením.
Vliv ancymidolu na buňky BY-2 byl velice podobný vlivu běžně používaných inhibitorů
syntézy celulózy (isoxabenu, DCB).
V malformovaných částech buněk BY-2 byla poškozena jednak orientace mikrotubulů a
jednak ukládání celulózních mikrofibril. Vzniku malformací nebylo možné zabránit stabilizací
mikrotubulů taxolem. Ancymidol také inhiboval biosyntézu celulózy v protoplastech buněk BY-2.
Ancymidol má tedy dvojí roli, je inhibitorem biosyntézy giberelinů v rostlinách a zároveň
působí na giberelinech zcela nezávisle jako jako inhibitor syntézy celulózy.
4. Úloha izoforem aktinu v somatické embryogenezi smrku
Somatická embryogeneze je proces vzniku embryí ze somatických, který odpovídá svými
vývojovými fázemi embryogenezi zygotické, a používá se jak ke studiu embryogeneze, tak k
množení různých užitkových rostlin (von Arnold et al., 2002). Je to velice komplexní proces,
ovlivňovaný mnoha endogenními i exogenními faktory, při kterém dochází k vývoji embrya
přesně lokalizovaným dělením, růstem, diferenciací a řízeným odumíráním buněk. Všechny tyto
procesy jsou velice úzce spjaty s dynamikou cytoskeletu. V řízení polarity buněk, dělení, růstu,
transportu i programované buněčné smrti hraje důležitou roli aktin (Smertenko et al., 2003). V
různých fázích vývoje embrya a rostliny se uplatňují různé izoformy aktinu i četné asociované
proteiny. Aktinové drogy, jakou je latrunkulin B (Morton et al., 2000), depolymerují aktin
v buňkách, ale mohou být i výborným nástrojem ke studiu embryogeneze, jejímu ovlivnění i
případnému komerčnímu využití. Jako materiál pro výzkum embryogeneze posloužila somatická
embrya smrku.
Aplikace nízkých dávek latrunkulinu B (50-100 nM) během maturace somatických embryí
způsobovala odumření buněk suspenzorů, zatímco buňky meristematických center ošetření
přežívaly. To naznačilo různou citlivost aktinu v těchto dvou typech buněk. Ošetření
latrunkulinem ve výsledku urychlilo vývoj dobře vyvinutých embryí a eliminovalo embrya
nedostatečně vyvinutá.
Byly izolovány čtyři smrkové geny pro aktin. Analýzou jejich exprese během maturace
byly odhaleny tři izoformy, které jsou exprimovány více v buňkách suspenzoru a jejichž exprese v
průběhu maturace klesá. Tento pokles byl významnější po ošetření latrunkulinem B.
Sekvenční analýza ukázala substituci aminokyselin v oblasti vazebného místa pro
latrunkulin B v jedné z izoforem aktinu specifických pro suspenzor, která může měnit vazebnou
afinitu izoformy k latrunkulinu.

Latrunkulin B urychloval zrání studovaných embryí smrku a pomáhal selektovat embrya
bez vývojových vad, a to přednostní degradací citlivého aktinu suspenzorů a urychlením odumření
suspenzoru.
5. Časné projevy toxického působení hliníku na rostliny
Kationty hliníku značně limitují růst rostlin na kyselých půdách (vzniklých přirozenou
acidifikací nebo v důsledku lidské činnosti). Reakce rostlin na toxické působení hliníku je
komplexní proces, kterého se účastní mnoho buněčných struktur a jehož výsledkem je velice
rychlá zástava růstu kořene. Vlivem hlinitých iontů a kyselého prostředí dochází ke změnám
vlastností buněčné stěny, dynamiky mikrotubulů nebo poškození membrán (Kochian et al., 2005).
Která z ovlivněných buněčných struktur je přímo zodpovědná za rychlou zástavu růstu kořene
není jasné. V této práci jsme se zaměřili na velmi časné symptomy toxického působení hliníku ve
vztahu k mikrotubulárnímu cytoskeletu a plazmatické membráně. Pro tento účel jsme použili
hydroponicky pěstované semenáčky Arabidopsis thaliana. Tyto semenáčky vykazovaly omezený
růst kořenů již v prvních minutách po ošetření hliníkem.
Měření fluidity na izolované plazmatické membráně ukázalo, že hliník způsobuje rychlou
rigidizaci membrány, která může být částečně odstraněna aplikací benzylalkoholu, membránového
fluidizéru. Aplikace benzylalkoholu na kořeny rostlin ošetřených hliníkem částečně obnovila
jejich růst.
Byla pozorována inhibice endocytózy v kořenech po 10 minutách a naprostá ztráta
endocytózy po 20 minutách působení hliníku.
Kortikální mikrotubuly byly hliníkem stabilizovány a naproti tomu působením nízkého pH
došlo k jejich rozpadu během 30 minut.
Při použití drog ovlivňujících endocytózu a dynamiku mikrotubulů bylo prokázáno, že
inhibice endocytózy vedla k zpomalení růstu kořenů, zatímco změna dynamiky mikrotubulů růst
kořenů v prvních 30 minutách působení neovlivňovala.
Uvedené výsledky ukazují, že hliník způsobuje v první fázi působení značné snížení
fluidity membrán a inhibici endocytózy. Oba tyto jevy vedou ve výsledku k rychlé inhibici růstu
kořenů.
6. Závěry
► Byla popsána schopnost proteinu ARPC2, podjednotky ARP2/3 komplexu, vazby na
mikrotubulární cytoskelet. Tyto výsledky společně s lokalizačními experimenty proteinů
ARPC2 a ARP2 naznačily možnost, že ARP2/3 komplex může být přímým
zprostředkovatelem interakce mezi mikrofilamenty a mikrotubuly v procesech spojených s

přestavbou mikrotubulárního cytoskeletu v koordinaci s růstem závislým na aktinu v
rostlinné buňce.
► Bylo prokázáno, že vliv růstového retardantu ancymidolu na vznik tvarových malformací
buněk BY-2 spočívá v jeho dosud nepopsaném inhibičním účinku na syntézu celulózy.
Mikrotubulární cytoskelet v tomto procesu hrál pasivní roli a nebyl za tvarové malformace
přímo zodpovědný.
► Bylo zjištěno, že během zrání somatických embryí smrku jsou buňky meristematické a
buňky suspenzorové odlišně citlivé vůči latrunkulinu B, a to zřejmě díky odlišnému složení
aktinových izoforem v těchto buňkách. Aplikace velmi nízkých koncentrací latrunkulinu B
tak vede k selektivní smrti buněk suspenzoru a následně k selekci kvalitních a zdravých
embryí.
► Toxické působení hliníku způsobuje rychlou zástavu růstu kořenů. Bylo zjištěno, že mezi
její bezprostřední příčiny patří rigidizace plazmatické membrány kořenových buněk a
zástava endocytózy. K časným efektům působení hliníku patří též stabilizace kortikálních
mikrotubulů, která nepřispívá k rychlé zástavě růstu kořenů, avšak zřejmě hraje důležitou
roli během pozdních efektů působení Al na morfologii kořene.

1. Introduction
Cytoskeleton is highly dynamic spatial network of proteins, which is an essential
component of eukaryotic cells. Cytoskeletal filaments, i.e. microfilaments and microtubules,
mediate the signal transduction, the flow of information and building material, the movement or
anchoring of proteins and organelles as well as the movement of whole cells and organisms.
Cytoskeleton takes part in the cell division, growth as well as tissues and organs development.
Functional cytoskeletal network is a subject of continuous reorganization. Controlled growth,
degradation and transport of cytoskeletal subunits are involved in all processes in living cells.
Associated proteins are required for proper function and organization of cytoskeletal filaments.
Cytoskeleton is a highly conserved structure, therefore, many its structural and functional aspects
apply for animal as well as plant cells. However, there are functions and structures specific for
plants or animals only.
Cytoskeleton is involved in the plant morphogenesis (for review see Mathur a Hülskamp,
2002). Plant cells are non-motile and the organization of their tissues and organs are given by the
strict control of oriented cell division and growth. Both microfilaments and microtubules are
involved in these processes. Further, cell morphogenesis and development of complex cell shapes
of specialized cells are controlled by the cytoskeleton (Mathur, 2006). During all of growth and
development processes, the interaction and coordination of involved cytoskeletal structures is
required. Proteins associated with the cytoskeleton participate in these processes (for review see
Petrášek a Schwarzerová, 2009).
Due to its important roles, the cytoskeleton is intensively studied. Modern biochemical,
microscopic and molecular methods contributed substantially to our understanding of the
cytoskeleton functions. Because of wide range of actions in which cytoskeleton takes part, there is
still lot of unanswered questions. In my work I attempted to contribute to elucidation of the role of
cytoskeleton and its associated proteins in several situations related to the plant cell and whole
plants morphogenesis. This thesis is divided into four independent chapters connected through the
role of the cytoskeleton in plant cell morphogenesis. Actin-microtubule interactions, the effect of
growth retardant ancymidol in context of cell wall formation, the role of actin in development of
Norway spruce somatic embryos, and root cells response to toxic influence of aluminium are
studied.

2. The role of ARP2/3 complex in actin - microtubule interaction
Microfilaments and microtubules form two independent but well cooperating networks in
cells. Molecular basis of their mutual interactions is not still elucidated. ARP2/3 is a conserved
protein complex associated with microfilaments, which consists of seven subunits (Goley a
Welch, 2006; Mathur, 2005). Primary function of ARP2/3 complex is connected with actin
dynamics, because the complex nucleates and branches actin. ARP2/3 complex, however, is
suspected to be involved in actin - microtubule interaction as well. In this work we used the
protein ARPC2, one of ARP2/3 complex subunits, to study the function and localization of the
plant ARP2/3 complex. Our results suggested that ARPC2 interacts with both actin and
microtubules.
Nucleotide sequence of NtARPC2 was obtained from BY-2 cells. NtARPC2 was further
used for protein isolation, antibody production and fusion with GFP.
Fusion protein GFP-APRC2 transiently overexpressed in BY-2 cells formed network of
filaments of unknown nature. These filaments partly colocalized with microtubules, however, not
with actin filaments.
Cosedimentation assay of ARPC2 with actin and microtubules in vitro showed that protein
ARPC2 was capable of direct binding to both actin filaments and microtubules.
Indirect immunofluorescence visualisation of ARPC2 and ARP2 subunit of ARP2/3
complex showed their localization in fine and characteristically organized dots, which decorated
actin filaments and colocalized with microtubules in BY-2 cells.
Our results suggest that besides actin nucleation activity of ARP2/3 complex, the complex
has also another function in actin-microtubule interaction. It is possible that ARP2/3 complex acts
as a cross-linking factor between actin and microtubules and ARPC2 is the component directly
responsible for binding to microtubules.
3. Cellulose synthesis inhibitory action of ancymidol impairs plant cell expansion.
Ancymidol is a plant growth retardant with inhibitory effect on gibberellins biosynthesis.
The main effect of ancymidol action is the reduction of plant elongation. Ancymidol induced cell
shape malformations of BY-2 cells, and this action was, however, gibberellins insensitive
(Boříková et al., 2003). In the study of ancymidol action we focused on the cell wall and the
cytoskeleton, because these structures are responsible for the shaping of plant cells. Cellulose, the
major component of cell walls, is synthesised on the cell surface by cellulose-synthase complexes
(Cosgrove, 2005). The orientation of deposited cellulose microfibrils is under the control of
cortical microtubules (Wasteneys a Fujita, 2006). To study gibberellin - independent mechanism

of ancymidol action we used BY-2 cells and tobacco Nicotiana benthamiana seedlings as an
experimental material.
Ancymidol in 10 -100 µM concentrations induced cell shape malformations of BY-2 cells.
The size and number of malformations as well as the cell viability decrease was concentration-
dependent. The viability of cells decreased with increasing rate of malformations occurrence in the
cell culture. Nevertheless, 1 mM ancymidol did not induce cell malformations but inhibited cell
elongation and prevented the formation of cell plate between newly divided cells. These effects of
ancymidol were independent on gibberellins. Cell shape malformations induced by ancymidol
resembled the effect of commonly used inhibitors of cellulose biosynthesis (isoxaben, DCB).
In malformed parts of BY-2 cells, orientation of cortical microtubules was changed and the
deposition of cellulose microfibrils was impaired. Taxol-mediated stabilisation of microtubules,
however, did not prevent the formation of cell shape malformation. In regenerating protoplast of
BY-2 cells, ancymidol inhibited cellulose regeneration.
Here we report the dual action of ancymidol. Ancymidol inhibits gibberellins biosynthesis
in plants. In higher concentrations, however, ancymidol inhibits cellulose synthesis.
4. The role of actin isoforms in somatic embryogenesis in Norway spruce
Somatic embryogenesis is defined as a process of embryos formation from somatic cells,
whose developmental stages correspond to zygotic embryogenesis. Somatic embryogenesis is
widely used for studying of embryogenesis and for vegetative propagation of plants as well (von
Arnold et al., 2002). Embryogenesis is a multi-step process influenced by number of endogenous
and exogenous factors, where embryo develops by the sequence of controlled cell division, cell
growth, cell differentiations and programmed cell death. Entirely all these processes are related to
the cytoskeletal dynamics, where especially actin filaments play important roles in the cell polarity
control and programmed cell death (Smertenko et al., 2003). Different actin isoforms and
associated proteins participate in various developmental stages of embryos development.
Latrunculin B is widely used as an anti-actin drug that depolymerises actin (Morton et al., 2000)
and thus represents a suitable tool for studies of embryogenesis. In this study we used somatic
embryos of Norway spruce as an experimental material for the research for the study of
embryogenesis.
Application of low doses of latrunculin B (50-100 nM) during somatic embryos maturation
killed predominantly suspensor cells while leaving cells in meristematic centres alive. This
indicated differential sensitivity of actin in these two cell types. The treatment resulted in faster
development of more advanced embryos and the elimination of insufficiently developed ones.

We isolated four spruce actin genes that encoded different actin isoforms and analysed
their expression during embryo maturation. We detected three actin isoforms, which were
expressed predominantly in suspensor cells and their expression decreased during maturation. This
expression decline was enhanced by latrunculin B treatment. Sequence analysis revealed amino
acid substitutions in latrunculin B-binding site in one of the suspensor-specific actin isoforms,
which may be responsible for its altered binding affinity to latrunculin B. We hypothesized that
this altered affinity of one of suspensor-specific actin isoforms may be responsible for higher
sensitivity of suspensor cells to latrunculin B treatment.
Latrunculin B blocked suspensor development, accelerates maturation of embryos and
selects high quality embryos.
5. Early effects of aluminium toxicity to plants
Aluminium toxicity is the main limiting factor for plant growth on acid soil. The toxic
influence of aluminium has probably multiple targets in root cells. Aluminium treatment results in
rapid cessation of root growth. Aluminium affects the cell wall, microtubule dynamics and plasma
membrane (Kochian et al., 2005). It is not well understood which of these structures is responsible
for rapid cessation of root growth. In this study we focused on early symptoms of aluminium
toxicity in microtubule cytoskeleton and plasma membrane. For this purpose we used hydroponic
cultivation of Arabidopsis thaliana seedlings.
The root growth was inhibited within first minutes of aluminium treatment. Measurements
of fluidity of plasma membranes isolated from Arabidopsis showed that aluminium caused rapid
rigidization of membranes. Application of membrane fluidizer benzyl alcohol was capable of
partial restoration of both membrane fluidity and the root growth. Further, we observed
endocytosis inhibition in roots affected by aluminium within 10 minutes and the loss of
endocytosis within 20 minutes of the treatment. Further we analysed the organization of cortical
microtubules in root cells and we observed that cortical microtubules were stabilized by
aluminium treatment within first 30 minutes and disrupted during prolonged exposition to
aluminium.
Using drugs affecting endocytosis and microtubule dynamics we showed that vesicle
trafficking, but not microtubule stabilization, inhibited root growth within first 30 minutes of
aluminium application. Therefore we
We concluded that aluminium-induced loss of membrane fluidity and endocytosis
inhibition were responsible for root growth inhibition within minutes of aluminium treatment.
Aluminium effect on microtubules played probably an important role as a late effect of aluminium
toxicity (within hours).

6. Conclusions
► The capability of ARPC2 protein (subunit of ARP2/3 complex) to bind microtubules was
described in this study. Our results indicated that ARP2/3 complex may mediate direct
interaction between microfilaments and microtubules in processes where the coordination of
microtubule reorganization and actin mediated growth was required.
► We showed that cell-malforming effect of growth retardant ancymidol was based on its
inhibitory action on cellulose synthesis. Microtubules played a passive role in this process
and were not directly responsible for changes of the cell shape induced by ancymidol.
► Meristematic and suspensors cells in maturating somatic embryos differed in sensitivity to
latrunculin B, probably due to different composition of actin isoforms in these cells.
Application of low doses of latrunculin B resulted in selective death of suspensor cells and
thus contributed to the development of high-quality embryos.
► Aluminium caused rapid cessation of root growth of Arabidopsis thaliana seedlings. We
showed that the immediate reason of root growth cessation was plasma membrane
rigidization and loss of endocytosis in root cells. Aluminium stabilized cortical microtubules
within minutes and induced their loss within hours of treatment. However, the effect of
aluminium on microtubules was not responsible for rapid root growth cessation, but
probably played an important role as a late effect of aluminium toxicity.

The co-ordination of cell wall synthesis with plant cell expansion is an important topic of
contemporary plant biology research. In studies of cell wall synthesis pathways, cellulose
synthesis inhibitors are broadly used. It is demonstrated here that ancymidol, known as a plant
growth retardant primarily affecting gibberellin biosynthesis, is also capable of inhibiting cellulose
synthesis. Its ability to inhibit cellulose synthesis is not related to its anti-gibberellin action and
possesses some unique features never previously observed when conventional cellulose synthesis
inhibitors were used. It is suggested that ancymidol targets the cell wall synthesis pathway at a
regulatory step where cell wall synthesis and cell expansion are coupled. The elucidation of the
ancymidol target in plant cells could potentially contribute to our understanding of cell wall
synthesis and cell expansion control.

Background: Somatic embryogenesis in spruce is a process of high importance for biotechnology,
yet it comprises of orchestrated series of events whose cellular and molecular details are not well
understood. In this study, we examined the role of actin cytoskeleton during somatic
embryogenesis in Norway spruce line AFO 541 by means of anti-actin drugs.
Results: Application of low doses (50-100 nM) of latrunculin B (Lat B) during the maturation of
somatic embryos predominantly killed suspensor cells while leaving the cells in meristematic
centres alive, indicating differential sensitivity of actin in the two cell types. The treatment
resulted in faster development of more advanced embryos into mature somatic embryos and
elimination of insufficiently developed ones. In searching for the cause of the differential actin
sensitivity of the two cell types, we analysed the composition of actin isoforms in the culture and
isolated four spruce actin genes. Analysis of their expression during embryo maturation revealed
that one actin isoform was expressed constitutively in both cell types, whereas three actin isoforms
were expressed predominantly in suspensor cells and their expression declined during the
maturation. The expression decline was greatly enhanced by Lat B treatment. Sequence analysis
revealed amino-acid substitutions in the Lat B-binding site in one of the suspensor specific actin
isoforms, which may result in a different binding affinity for Lat B.
Conclusions: We show that manipulating actin in specific cell types in somatic embryos using Lat
B treatment accelerated and even synchronized the development of somatic embryos and may be
of practical use in biotechnology.

Aluminum-induced root growth cessation is associated with loss of membrane fluidity
and endocytosis inhibition

Aluminum (Al) toxicity is the main limiting factor in crop production on acid soils. The main
symptom of Al toxicity is a rapid inhibition of root growth, but the mechanism of root growth
cessation remains unclear. Here we examined the earliest changes in the plasma membrane,
endocytosis, and cortical microtubule organization in the root tip cells of Arabidopsis thaliana. Al
suppressed root growth within 2 minutes, inhibited endocytosis within 10 minutes of exposure and
stabilized cortical microtubules within the first 30 minutes. Spectrofluorometric measurements of
the plasma membrane isolated from Arabidopsis plants and labeled with the fluorescent probe
laurdan showed that Al induced a reduction in membrane fluidity. Application of the membrane
fluidizer, benzyl alcohol, restored partially membrane fluidity and also partially restored root
growth during first 30 minutes of Al treatment. Using the inhibitor of vesicle trafficking brefeldin
A and microtubule-stabilizing compound taxol we showed that vesicle trafficking, but not
microtubule stabilization inhibits root growth within first 30 minutes after application. We
concluded that Al-induced loss of membrane fluidity and endocytosis inhibition occurred very
early during Al toxicity in plant roots and were responsible for root growth inhibition within
minutes of Al treatment.
