PCR 을 이용한 virus 검사항목• HPV (16,18)• Polioma Virus (BK,J
C) - RFLP• CMV• Parvovirus• HSV type 1,2• HHV type 6

Virus 검사에서 검체의 채취
• 호흡기 검체 - nasopharyngeal aspirate
• 대변 - 발병 후 4 일 이내에 채취• 혈액 - sodium citrate tube
• 뇌척수액 - 혈액이 섞이지 않게• 면봉검체 - 수포성 :26G 주사기• 소변 - 중간뇨 10 mL
• 기타검체 - 마르기쉬운 검체 : 운송배지
DNA 분리방법• Lysis: 0.5% SDS, Proteinase K
• Purification of nucleic acid : Extraction with Phenol:Chloroform:Isoamyl alcohol
• Concentrating Nucleic acid: Ethanol precipitation, sodium acetate
• DNA washing
• DNA solubulization
RNA 분리방법• Lysis:Tri-reagent
• Phase separation: chloroforum
• RNA precipitation: isopropyl alcohol, Glycogen
• RNA washing
• RNA solubulization
Coprecipitants
• During centrifugation, carriers generate a visible pellet that facilitates handling of the target nucleic acid

Detection
• Sensitivity of detection with ethidium bromide – 10 ng of DNA 가 검출될 수 있다 .
• 기타 염색에서의 검출율 Polyester-backed polyacrylamide gel 에서는
silver stainning 을 하면 picogram 까지 검출할 수 있다 .
Quantitation of nucleic acids
• Absorbance at 260 nm by spectrophotometer
• Emission at 458 nm in the presence of Hoechst 33258 by fluorometer
• Dipstick (a kit from Invitrogen)
• Ethidium bromide spot test by
UV transilluminator
Staining nucleic acids
• Ethidium bromide- staining DNA in gels(~10 ng/band)
• Methyene blue
• Silver staining
- < 1.0 ng of DNA
analyze single-stranded DNA by SSCP
• SYBR dyes
Summary
• Labor intensive 하지만 bcr-abl gene rearrangement 검사와 같이 1 년에 몇 case되지 않는 경우 기존 기구를 이용하여 setting 할 수 있겠습니다 .
• Nucleic acid 분리방법의 standard method를 이해함으로써 상품화된 시약을 평가 할 수 있겠습니다 .
