HPLC Columns Introduction

Choosing the Right HPLC Column1. Set Your Separation Goals. Determine if your application requires high resolution, short analysis time,

maximum sensitivity, long column life, low operating cost, or other important factors.

2. Choose Your Packing Material. Read below for an explanation of different specifications and how they affect your separation. Alltech’s key column families are described on page 128.

3. Choose Your Column Format. See pages 126-127 for a summary of column formats and how they affect your separation.

Packing Materials

Understanding the specifications of HPLC packing materials and how they affect a separation will help you narrow your column choices. You will find all of these specifications for Alltech’s HPLC packings on pages 129-130.

Base Material

Silica-based packings are physically strong and will not shrink or swell. They are compatible with a broad range of polar and non-polar solvents. Most silica phases are stable from pH 2-8, but newer phases, such as Alltima™ HP, are stable from pH 1-10. Silica provides sharp peaks with small molecules.

Polymer-based packings, such as methacrylate, are compressible and may shrink or swell with certain solvents. Many polymers are stable from pH 1-14. Because of this expanded pH range, most polymeric columns can be thoroughly cleaned with strong acids or bases to extend column life.

Particle Size

Smaller particle sizes give higher efficiency and higher resolution than larger particle sizes. However, larger particle sizes offer faster flow rates and lower back-pressure. Typical particle sizes range from 3µm to 20µm, and new 1.5µm particle sizes are available to maximize resolution on short columns. A 5µm particle size represents the best compromise between efficiency and back-pressure.

Particle Shape

Most modern HPLC packings have spherical particles, but some are irregular in shape. Irregular particles have larger surface areas, and are relatively inexpensive. Spherical particles offer lower back-pressure, and higher performance, stability, and reproducibility than irregular particles.

Endcapping

Silica-based reversed-phase packings have free silanol groups that will interact with polar compounds. Endcapping the bonded phase minimizes these secondary interactions.

Choose endcapped phases if you do not want interactions with polar compounds. Choose non-endcapped phases if you want enhanced polar selectivity, for stronger retention of polar organic compounds.

Carbon Load

For silica-based reversed-phase packings, carbon load indicates the amount of functional bonded phase attached to the base material. Phases with lower carbon loads are more weakly hydrophobic, which may significantly reduce retention times over phases with higher carbon loads. However, a higher carbon load will give higher capacity and often greater resolution, especially for compounds of similar hydrophobicity.

Pore Size

Choose a pore large enough to completely enclose your target molecule. If your molecule is larger than the pore, it will be difficult or impossible to retain. In general, packing materials with a smaller pore size have higher surface areas and higher capacities than packing materials with larger pore sizes. Larger pores are better for interaction with large compounds, such as proteins.

Surface Area

This is the available surface, most of which is within the pores, for interaction with the sample. A larger surface area typically indicates a greater number of pores, and therefore a higher overall capacity. Smaller surface areas equilibrate faster, which is important for gradient elution analyses.

Phase Type

Polymerically bonded phases have functional chains bound to the base silica particle at multiple attachment points and can involve cross-linking between chains, while monomerically bonded phases have a single attachment point. Historically, polymeric packings have resulted in better column stability under pH extremes. However, new high-purity silica phases are very stable whether monomerically or polymerically bonded. Monomerically bonded phases generally offer rapid mass transfer and high column efficiency to better resolve chemically similar analytes.

Choosing the Right HPLC Column Formats

Column Length

10-50mm length columns Rapid separations, work best with 3µm particle sizes.

100-150mm length columns Standard separations, work best with 3-5µm particle sizes.

≥250mm length columns High-resolution separations, work best with 5-10µm particle sizes.

Column i.d.

Narrower columns provide better mass sensitivity, accommodate smaller sample sizes, and significantly reduce solvent usage. Wider columns allow for larger sample sizes and minimize the negative effects of your system’s dead volume.

2.1mm i.d. columns work best with a microbore flow cell and an internal loop injector, but they can be used with standard analytical hardware if you can tolerate an approximate 20% loss in efficiency/resolution and an increase in tailing.

Column i.d.

0.3 to 1.0mm i.d. columns For high sensitivity or limited sample volumes. Use with LC/MS instrumentation.

2.1mm i.d. columns For high sensitivity or limited sample volumes. Use with low dead-volume instrumentation.

3.0mm i.d. columns For solvent reduction. Use with standard HPLC instrumentation. Compatible with both APCI and ESI.

4.0 to 4.6mm i.d. columns For standard separations. Use with standard HPLC instrumentation.

7-10mm i.d. columns For small-scale preparative separations.

22mm i.d. columns For large-scale preparative separations.

Column Length

For the same packing material, shorter columns provide faster run times, while longer columns provide better resolution. Consider the complexity of your sample and your desired run time to find the best column length for your application.

In general, choose smaller particle size packings in shorter column lengths to achieve good resolution in shorter run times. Highly complex samples may require longer columns, even with small particle size packings, to efficiently separate all of the sample components.

What are Waters-style endfi ttings?

Column endfi ttings accept connecting tubing from the injector or detector. To avoid leaks or excessive dead volume, the column endfi tting must have a stop depth that matches the stop depth of the connecting tubing. Waters Corporation makes endfi ttings with a different stop depth than that of most other column manufacturers. Alltech offers column endfi ttings made with either the industry standard stop depth or with Waters stop depth.

However, stop depth is only critical when using stainless steel ferrules, because stainless steel ferrules swage onto the connecting tubing and permanently fi x the stop depth. What is hydrophobic interaction chromatography?

Hydrophobic interaction chromatography separates compounds based on the interaction between hydrophobic groups on a

protein and a hydrophobic ligand on the column packing. It is generally run in an all-aqueous environment using a reverse salt gradient (high to low salt concentration). A number of factors affect protein binding to the HIC packing such as the type of ligand, ligand density, the base material, degree of protein hydrophobicity, and the type of mobile phase salt used.

RP-DVBRP-DVB is a reversed-phase non-polar packing with a high degree of aromatic character. SM-500, with a 500Å pore, is recommended for most applications. LM-1000, with a 1000Å pore, is ideal for molecules 100,000 Daltons or larger.

C18-DVBC18-DVB has C18 chains bonded to the DVB base. These columns are more hydrophobic than traditional C18 columns and have different selectivities due to the absence of silanols.

Polyamine-DVBPolyamine-DVB is a normal-phase column for simple sugar and polysaccharide applications. The DVB-based packing provides a non-reactive amine surface that lasts longer and equilibrates faster than traditional silica-based amine columns. Polyamine-DVB columns can be used with aqueous mobile phases, even 1M NaOH. Ideal for carbohydrate analyses requiring sharp peaks and short run times.

When should I use ZirChrom® EZ or MS columns?The new deactivated ZirChrom® reversed-phase columns are best suited for applications requiring the unique selectivity of zirconia but limited to conventional reversed-phase operating conditions. secondary amino groups for thermal and chemical stability. PAC columns separate primarily by reversed phase or weak anion exchange with fast equilibration.

Jordi Polyamine ColumnsPolyamine-DVB is a normal-phase column for simple

sugar and polysaccharide applications. The DVB-based packing provides a non-reactive amine surface that lasts longer and equilibrates faster than traditional silica-based amine columns. Polyamine-DVB columns can be used with aqueous mobile phases, even 1M NaOH. Ideal for carbohydrate analyses requiring sharp peaks and short run times.
