DANIEL VANEGAS ISAZATATIANA ZAPATA CORREA
INTRODUCTION
HELICOBACTER
• Gram-negative bacteria.
• Helix shape.
• Microaerobic conditions at 37°C.
• Doesn´t need hydrogen.
INTRODUCTION
HELICOBACTER PYLORI
• Lives on the gastric epithelium.
• Damage the mucus layer gastric acid.
• Ulceration and possibly gastric cancer.
• L- asparaginase.
INTRODUCTION
L-ASPARAGINASE
• Destroys asparagine outside the cells.
• Only intacellular reserve.
• Proliferation increases the asparagine demand.
INTRODUCTION
L-ASPARAGINASE – CELL CYCLE
• Relation l-asparaginase/ asparagine synthetase.
• Inhibition of cell cycle.
• Fibroblastic and gastric cell.
GENERAL OBJETIVE
Demonstrate that H. pylori L-asparaginase plays an important role in cell-cycle inhibition induced by BCF on cultured cells, and show that this inhibition is more pronounced in cells with low expression levels of the enzyme asparagine synthetase

DISCUSSION
AUTHOR WHAT HE SAID YES OR NOT
Sommi P, et al.
Following exposure to H. pylori BCF, HDF, MKN7 and MKN28 cells showed a significant increase in the number of cells in G1 phase, and, concomitantly, a decrease in those in the S phase.
Yes
Ding Y, et al.
In fact, the cell-cycle block of cell clones, especially leukemic, exposed to this enzyme class hasbeen previously described to depend on the expression level ofasparagine synthetase
Yes
DISCUSSION
AUTHOR WHAT HE SAID YES OR NOT
Fiocca R, et al.
The observation that the protein has a predicted signal peptide, suggests that protein transit into the external environment might occur through secretion or through release of outer membrane vesicles
Yes
Keating MJ, et al.The activity of the recombinant enzyme steadily increases along the pH gradient observed throughout the gastric mucus layer
Yes
CONCLUSIONS
• Helicobacter pylori affects 50% of world people, for this reason its important to know about it and identify some diagnostic method that helps in a early detection.
• This study is important because, it provides information on the behavior of H. pylori and that information can be used to studies to identify possible treatments.
CONCLUSIONS
• L-asparaginase is the responsible factor for cell-cycle inhibition of gastric cell lines for this reason it could be used on the diagnostic of H. pylori-related diseases.
• The L-asparaginase’s role as a potential pathogenic factor could be thoroughly analysed by studying its internalisation in cells of infected patients.
