Over 40 % av alle
publikasjonene i 2010 var i nivå 2 journaler.
Denne trenden er videreført i 2011. Dette er
absolutt noe å være stolte av!
Vi gleder oss over at Thomas Arnesen har fått
formalisert sin stipendkontrakt med BFS og at
HelseVest fant midler til å støtte PhD-stipend til
prosjektet hans. Flere tildelinger til grupper ved
MBI fra Kreftforeningen gleder også. Gaver til
forskningen blir mer og mer viktig for oss og da
får vi heller ta utfordringen med at slike gaver ikke
passer inn i universitetets økonomiske
driftsmodell.

instituttet.

Fish based diets have been linked to the amelioration of methylmercury (MeHg) induced symptoms inseveral epidemiological studies, particularly due to their contents of marine n−3 fatty acids. It has beensuggested that n−3 fatty acids may mask the detrimental effects of MeHg due to their beneficial effecton the same biological functions which are negatively affected by MeHg. However, in vitro studies haveimplied that there may be direct interactions between the marine n−3 FAs and MeHg, which amelioratesMeHg toxicity through interactions at a biological level. To understand how marine n−3 FAs and MeHginteract in fish as a biological system, we wanted to investigate molecular interaction in a fish cell system.
Atlantic salmon kidney (ASK) cells were pre-incubated with the marine n−3 FAs docosahexaenoic acid(22:6n−3, DHA) and eicosapentaenoic acid (20:5n−3, EPA) before exposing them to MeHg. Modulatingeffects of the marine FAs on MeHg toxicity were subsequently assessed using the exploratory techniqueof proteomics, in a factorial design.
Thirty-four differentially regulated proteins were identified. From these; twenty-seven were shown tobe differentially regulated by MeHg, twelve were regulated by the fatty acids, and another eight showedinteraction effects between MeHg and the FAs. Several of the proteins were concomitantly affected byMeHg- and FA-main effects, as well as interaction effects. Functional annotations and pathway analysisof the proteins revealed that marine n−3 FAs and MeHg concurrently affected the abundance of pro-tein markers relating to such molecular mechanisms as: cell signaling, calcium homeostasis, structuralintegrity, apoptosis, and energy metabolism.
In conclusion, both marine n−3 FAs and MeHg can differentially affect the abundances of the sameproteins, indicating modulating effects of EPA and DHA on MeHg metabolism, and possibly on its toxicity.

1. Introduction
The marine n−3 fatty acids (FA) docosahexaenoic acid (22:6n−3;DHA) and eicosapentaenoic acid (20:5n−3; EPA) have proven to beof great importance in a healthy balanced diet (Calder et al., 2010).These essential FAs have shown beneficial effects on human healthsuch as: onset of coronary heart disease (Mozaffarian and Rimm,2006), pregnancy and infant development (Cetin and Koletzko,2008), cognitive health (Albanese et al., 2009; Hibbeln et al., 2007)and immune regulation (Calder, 2007). The source of marine n−3FAs for human consumption is mainly through a seafood based

diet, which unfortunately is prone to also contain environmen-tal contaminants. This has led to a debate whether the beneficialeffects of nutrients from seafood outweighs the harmful effects ofpossible toxicants (Bushkin-Bedient and Carpenter, 2010; Strömet al., 2011). There is however, another aspect of co-occurring nutri-ents with contaminants, specifically, they may directly interactand possibly alter the toxicology of the contaminant. Lately, stud-ies have emerged investigating the interactive effect of nutrientsand contaminants, demonstrating that selenium can ameliorateMeHg toxicity (Rooney, 2007), and marine n−3 FA oxidation prod-ucts protect endothelial cells against coplanar PCBs (Majkova et al.,2011). The mechanisms responsible for nutrient-mediated protec-tion against contaminants are also assessed, as for example theimportance of sufficient zinc levels in maintaining expression ofdetoxifying enzymes (Shen et al., 2008). Overall, the increasedinterest in nutrient effects on toxicants has emerged as a valuable

Atlantic salmon (Salmo salar) and brown trout (Salmo trutta) possess two distinct subpopulations of IgMwhich can be separated by anion exchange chromatography. Accordingly, there are two isotypic genesin these species, related to ancestral tetraploidy. In the present work it was verified by mass spectrometrythat IgM of peak 1 (subpopulation 1) have heavy chains previously designated as B type whereas IgMof peak 2 (subpopulation 2) have heavy chains of A type. Two adjacent cysteine residues are presentnear the C-terminal part of B, in contrast to one cysteine residue in A. Salmon IgM of both peak 1 andpeak 2 contain light chains of the two most common isotypes: IgL1 and IgL3. In contrast to salmon andbrown trout, IgM of rainbow trout (Oncorhynchus mykiss) is eluted in a single peak when subjected toanion exchange chromatography. Surprisingly, a monoclonal antibody MAb4C10 against rainbow troutIgM, reacted with A in salmon, whereas in brown trout it reacted with B. It is plausible to assumethat DNA has been exchanged between the paralogous A and B loci during evolution while maintainingthe two sub-variants, with and without the extra cysteine. MAb4C10 was conjugated to magnetic beadsand used to separate cells, demonstrating that transcripts residing from captured cells were primarilyof A type in salmon and B type in brown trout. An analysis of amino acid substitutions in A and B ofsalmon and brown trout indicated that the third constant domain is essential for MAb4C10 binding. Thiswas supported by 3D modeling and was finally verified by studies of MAb4C10 reactivity with a series ofrecombinant 3 constructs.

1. Introduction
IgM is the primary systemic antibody in teleost fish. Teleost IgMis typically a tetramer (Acton et al., 1971), and each monomer con-sists of two identical heavy chains and two identical light chains. The heavy chain () of secreted IgM consists of one variable Igdomain and four constant Ig domains (1, 2, 3 and 4). membrane anchored form of IgM, i.e., the B-cell receptor, is oneIg domain shorter than the secreted form as a result of a specialsplicing pattern in teleosts which excludes 4 (Ross et al., 1998).
A J-chain homolog has been revealed in representatives of allvertebrates except cyclostomes and bony fish (Klimovich et al.,2008). Thus, presence of a J-chain appears to correlate with theability to form IgM pentamers; in mammals, amphibians, reptilesand cartilaginous fishes.
Purification of serum IgM from salmonid fish is usually per-formed by a combination of anion exchange chromatography andgel filtration, or by affinity chromatography employing specificantibodies against the IgM of interest (Kobayashi et al., 1982;Haavarstein et al., 1988; Fuda et al., 1991; Sanchez et al., 1993,1995; Magnadóttir et al., 1996, 1997).
