[Abstract] Objective Toevaluatetheeffectsofthestaticmagneticfield(SMF)onbonemarrowmesenchymalstemcells(MSCs)osteogenicdifferentiationandexplorethepossiblemechanism.Methods Densitygradientcentrifugationisolatedratbonemarrowmononuclearcellswereculturedinvitro.MSCsofthe3rdand4thpassageswerechosenastargetcells.Afterosteogenicinduction,theMSCswereexposedtoSMFat0,0.5,1.5,2or3mT.Theactivityofalkalinephosphatase(ALP)wasassayedwithIFCC.ThecalciumnodesofMSCswerestainedbyalizarinred.Theconcentrationsofosteocalcin(OCN)andtypeⅠ collagenweremeasuredbyradioimmunoassay.WesternblottingwasusedtodetectthephosphorylationofMAPKP38.SpecificP38inhibitorSB253580wasaddedintotheculturemediaofMSCstoinvestigatetheroleofP38inthisprocess.Results SMF,especiallyat1.5and2mT,increasedtheactivityofALP,thenumberofcalciumnodesandtheconcentrationsofOCNandtypeⅠ collageninMSCs.SMFactivatedMAPKP38signalingmolecule.BlockingtheP38signalpathwaybySB253580partiallyreversedtheeffectsofSMFonosteogenicdifferentiationofMSCs.Conclusion SMFpromotestheosteogenicdifferentiationofMSCsandtheP38MAPKpathwayisinvolvedinthisprocess.
