Chemical structure and Shwartzman activity of Lipid A

Abstract
Lipopolysaccharide (LPS) is a structural component in the outer membrane of the cell wall of
Gram-negative bacteria, which mainly consists of O-specific polysaccharide, core oligosaccharide
and lipid A moieties. The lipid A region of LPS is responsible for most of its endotoxic and
biological activities. We examined the relationship between the chemical structure of lipid A
and local Shwartzman reactivity by using the purified lipid A components from five bacterial
strains. The local Shwartzman reaction can be partially expressed by different lipid A structures
in both hydrophilic backbone and fatty acyl residues; however, when the structures have a same
backbone the potency varied markedly depending on the structure of the acyl residues.
