Phn 1: BACILLUS CEREUS1. Gii thiu Mt bo co trc y v v ng c thc phm vi vi khun Bacillus spp. c thc hin vo nm 1906 khi Lubenau m t mt t bng pht trong mt vin iu dng ni 300 bnh nhn v nhn vin mc cc triu chng ca bnh tiu chy, au bng v nn ma. 2. Hnh thi v c tnh Cc Bacillus l vi khun _Gram dng

3. Cc triu chng v bnh lm sn: Vi khun Bacillus cereus phn b nhiu trong t nhin, nhim vo cc loi thc n qua m hay tr lnh lu, thng gy ng c thc phm.c t:vi khun sn sinh 2 loi c t c t gy tiu chy (Type 1): Diarrhoed toxin. Vi khun sn sinh c t trn tht , rauqu, gia v. Bn cht l mt loi protein gy hy hoi biu b v nim mc rut gy tiuchy c th nguy him n tnh mng. c t gy nn ma (Type 2): emetic toxin. Vi khun nhim trong go, cm ngui, ucc loi. 4. Phn lp v nhn dng4.1 Truyn thng: Nhm cc phng php ny da trn c im pht trin ca vi sinh vt trn cc mitrng c trng v c im sinh l, sinh ho ca cc chng, cc loi vi sinh vt khc nhau.u im:Thao tc n gin, d lm, khng phi u t dng c, thit b t tin. Nhc im:- nhy khng cao.- Yu cu k nng thao tc cao.-Tn nhiu nhn cng.- Thi gian phn tch thng ko di. 4.1.1 nh lng B.cereus bng phng php m khun lc:

Tin hnh th nghim sinh ha

Do c hnh thi c trng trn cc mi trng thch chn lc nh: MannitoEgg-Yolk-Polymycin(MYP),CereusSelectiveAgar(MOSSEL),PolymicinElgelbMannitolBromothymol Blue Agar (PEMBA), nn B.cereus cn c pht hin v nh lng bng mi trng ny.

B. cereus trn thch MYPPhn ng sinh ha: Th nghim ln men glucose:Cy vi khun vo 3ml canh Phenol Red Glucose Broth 350C/ 24h, k kh lc mnh ng nghim,quan st c (s pht trin). Nu mi trng t sang vng th chng t c s sinh acid glucose trong iu kin k kh.

Kt qu th nghim ln men glucose Th nghim kh nng kh nitrate:Cy vi khun vo 5ml canh trng Nitrate Broth 350C/24h, b sung vi git thuc th nitrate, c mu cam xut hin trong 10 pht th chng t nitrate b kh thnh nitrit.

ng B v C c b sung thuc th alpha-naphthylamine v sulfanilic acid. ng B cho kt qu dng tnh (nitrate b kh thnh nitrite ), ng C cho kt qu m tnhvi nitrite. Th nghim VP:Cy vi khun trong mi trng MR-VP 48h, nhit 370C b sung thuc th (-Napthol) vo mi trng, lc nh. C 2 loi thuc th VP thch hp trong trng hp ny l:Thuc th Barritt:gm dung dch A l 5%-naphthol trong cn tuyti, dung dch B l 40% KOH hoc NaOH.Thuc th Koblentz: gmdung dch A l5%-naphthol trong cn 95%, dung dchB l 0.3% creatine, 40% KOH hoc NaOH.Khi s dng cc loi thuc th 2 thnh phn, trc tin nh 6 git dung dch A, sau nh 2 git dung dch B, b sung 1ml thuc th vo ng mi trng. Lc nh ng 1 pht oxi ha acetoin. c kt qu sau 20 pht, chm nht l 4h To phc hng l (+).

Th nghim kh nng thy phn tyrosine:Cy vi khun vo thch nghing tyrosine, 350C/48h xut hin khong trng xung quanh khun lc (chng t tyrosine b phn hy). Th nghim khng Lysozyme:Cy vi khun vo mi trng Nutrient Broth cha 0.001% lysozyme, thc hin tng t vi mi trng khng cha lysozyme, 350C/24h kim tra s tng trng trong 2 mi trng. Nhng ng c kt qu m tnh thm 24h kt lun vi khun c khng lysozme hay khng. Th nghim lm tan mu:Cy chun ln mi trng thch mu Tripticase Soy. 250C trong 24h. B.cereus lm tan mu mnh v to vng tan mu hon ton () 2-4mm xung quanh vng pht trin. B.thuringiensis v B.mycodes cng lm tan mu . B.anthracis thng khng tan trong mu sau 24h.

Sau kim tra s pht trin ca r gi (B.cereus khng to cu trc r gi, thng to nhm khun lc x x khc vi cu trc r gi c trng ca B.mycoides (cu trc ging nh r hoc tc m rng vi centimet t v tr cy).

4.2.1.2 Phng php Elisa gin tip (Indirect Elisa)Khng nguyn s c cho vo y ging. Khng nguyn s lin kt c nh vo b mt ca y ging. mt thi gian ngn, sau tin hnh ra tri. Nhng khng nguyn no khng lin kt s c a ra ngoi.a khng th mc tiu c hiu vo ging, mt thi gian v tin hnh ra tri, ch nhng khng th c hiu mi c gi li cng khng nguyn trn b mt y ging.a tip dung dch m cha khng th th cp c gn emzyme vo, khng th ny s lin kt vi khng th mc tiu. To phc hp khng nguyn_khng th mc tiu_khng th th cp_E. Cng tin hnh v ra tri.Khi cho dung dch cha c cht vo ging, c cht di xc tc ca E s c phn ng to mu.Phng php ny ch khc vi dng trc tip l phi s dng khng th th cp gn E do khng th mc tiu khng th gn vi E.

4.2.1.3 Phng php Elisa Sandwich:Trc tipMu cn phn tch c cho v trong a ging c ph khng th chuyn bit. Khng nguyn trong mu s kt hp vi khng th trong a ging v tr nn bt ng. a ging ny tip tc c ra loi b nhng khng nguyn khng kt hp c vi khng th. Sau , phc hp khng th - enzym (gm khng th lin kt vi enzym qua lin kt cng ha tr) c thm v. Khng th ca phc hp ny s kt hp vi khng nguyn bt ng trong a ging cn li sau khi ra hnh thnh 1 bnh sandwich gm: khng th khng nguyn khng th/enzyme trong a ging.Sau khi loi b khng nguyn hoc khng th, cc cng hp t do, c cht v cht pht mu tng ng vi enzyme s c cho v v mu s xut hin trong cc ging th. Vic nh lng khng th hoc khng nguyn c thc hin bng cch o mt quang gia mu v cht chun nh my c ELISA.

Xc nh Bacillus Cereus Bng Phng Php ElisaHai loi c t gy ra trng c thc phm do Bacillus cereus sinh ra l Emetic Toxin (c t gy nn) v Diarrhoeal Enterotoxin (c t gy tiu chy). Phng php xc nh Bacillus Diarrhoeal Enterotoxin (BDE) bng k thut phn tch Elisa c Terca International Pty a ra qua b Kit Tecra Bacillus Diarrhoeal Enterotoxin Visual Immuno Assay.4.2.2 Phng php PCRK thut PCR (Polymerase Chain Reaction) do Kary Mullis (Nobel 1993) pht minh, l mt k thut hin ph bin trong sinh hc phn t, da trn phn ng khuych i gene. Cc cy sn non cha vi sinh vt axenic t do thu c bng cch nui cy m. Ba bnh cha 400 ml dung dch Murashige & Skoog (Murashige & Skoog, 1962) v ba bnh vi nc ct tit trng c chun b. Cy sn c chuyn qua cc bnh v c tim chng 108 CFU ml-1 ca B. pumilus MAIIIM4a. Mu cha cy sn khng tim c kim sot. Sau 5 ngy cc cy sn c tch thnh thn, r v l cy v mi m c ct thnh on khong 2 cm2. Mi phn m sn c c nh trong mt gii php Karnovksy ciu chnh (Kitajima & Leite, 1999) v cc mu c gi mc 8C qua m. Cc mu c ra 3 ln vi m cacodylate (0,05M) trong 10 pht v c ngm trong 1% OsO4 (trong 0,05 M m cacodylate, pH 7.2) trong 1 gi. Cc mnh sn c ra nm ln, 15 pht i nc ct mt ln. Cc mu b loi nc trong mt dy ethanol (30, 50, 70, 90 v 100%), c 15 pht thay i mt tng nng ethanol mt ln. Lp i lp li ba ln. Qua 24h, 48h, 72h ly ra b i tm du hiu bnh khoai ty.Trng hp kim tra ln u (24h) nu c du hin bnh khoai ty, chng t bt m b nhim nng.Nu kim tra ln th hai (48h) thy du hiu bnh khoai ty, bt m b nhim trung bnh.V nu kim tra ln th ba (72h) mi thy du hiu bnh khoai ty, th bt m coi nh bnh thng.Bt m c trc khun khoai ty th khng nn dng sn xut bnh m, m nn dng vo ch bin cc thc phm khc nh m si.Trng hp cn phi s dng bt m lm bnh m (v cha c loi bt m tt khc), th bnh m phi nn nh, trng lng mi chic bnh khng qu 200gram.Bnh m sau khi ly l ra phi ngui ngay v phi s dng trong 24h.Nu hp thu m do bo qun khng tt bt m s b vn cc, mt s vsv bo t c kh nng s dng ng glucose (Bacillus mesentericus) s pht trin.
