Shih-Chen Chang Ph.D.2015 CBA President

The Chinese Bioscience Association (CBA), a bi-lingual and non-profit organization, has been established for more than 15 years in the Bay Area. Each year, CBA holds scientific conferences and workshops highlighting emerging trends and cutting-edge knowledge advancement in the life sciences associated topics, and has become one of the key platforms in promoting education, networking, and community building for life science professionals among students, post-docs, scientists, entrepreneurs, business community as well as academic institutes.

In 2015, CBA has organized several exciting activities providing avenues for CBA members and friends to socialize and network. With our close tie to the Asia Pacific regions as well as the Bay Area, one of CBA’s missions is to bridge the investments, industries and talents gaps between US and Asia. To this end, we have organized a well received workshop entitled “Modern Seafarers: Navigating Life Sciences Investments & between US and the Far East”, showcasing esteemed speakers from US, India, China and Taiwan sharing invaluable insights as well as current underlying challenges and opportunities in life science business investments in these regions. In addition, we also hosted The Singapore-Sichuan Hi-Tech Innovation Park delegation, and facilited meetings with company executives and entrepreneurs in the Bay Area, as well as provide opportunities to showcase the biomedical sciences innovation park jointly developed by Singapore and Sichuan, located in Chengdu, China. CBA will continue our support in the launch of the Ho Family Scholarship. And this year, we are pleased to announce that there will be two scholarships in lieu of the single award last year. Finally, we have another great turnout of over 150 at our annual summer picnic, providing an excellent opportunity for CBA members and their families and friends to socialize and network, amidst good music and great food.

The theme of this year’s conference is “Future of the Life Science Industry – Innovations and Opportunities”. The Bay Area is not only a fertile ground for developing novel bioscience technology in the United States, it also continues to house one of the world’s largest and most diverse biotech communities. To remain competitive, the Life Science industry must continue to develop and maintain innovative disease pipelines. It must also cut cost by streamlining clinical trials and optimize operational efficiency as well as forming win-win partnerships bringing about next generation technologies and access to the global market. In our 2015 annual conference, several aspects within the process from drug discovery to clinical use will be showcased, highlighting brilliant innovations and new opportunities that lie within.

Finally, I would like to thank all our sponsors, speakers, and conference/event participants. Your support is the key motivator that enables CBA to grow and improve. Last but not least, as a volunteer-based organization, I would like to personally thank the tremendous support from CBA board members, advisors, and volunteers. Without their dedication and tireless efforts, we would not have accomplished all that we have achieved thus far!

Sincerely,

President’s Message

ABSTRACTHighly massively parallel DNA sequencing using genomic nanoarrays continues to advance our ability to efficiently read DNA. Recent improvements in making patterned DNA Nano-Ball arrays (DNB arrays) provide easy use for all sample types and strong signal for long unbiased reads. These patterned nanoarrays combined with novel powerful cameras allow to generate sequence data with extreme efficiency (e.g. 50x WGS from 30 people per day per instrument) enabling whole genome sequencing (WGS) to be applied as the single comprehensive genetic test. Improved DNB array and fast imaging also enable cost effective desktop sequencers for various research and clinical “omics” application such as NIPT, PGS or targeted cancer tests. When Long Fragment Read (LFR) technology employing co-barcoded reads from long DNA fragments and 100xread coverage is used. Individual human genomes are sequenced at maximal sensitivity, including separate assembly of parental chromosome sequences termed haplotping or phasing. Furthermore, using co-barcoded reads, accurate WGS including detection and phasing of de-novo mutations (~1 error per Gb, ~6 errors per genome) is now achievable from ~10 cells such as in small biopsies obtained from 5-day IVF embryos and potentially fetal cells collected in early pregnancies. In addition, we show that using as few as five CTCs we can accurately find somatic mutations and gene deletions from the founding lineage of the metastatic breast cancer (paper under review). LFR’s co-barcoded reads also allow reference unbiased de-novo assembly of individual genomes for a more comprehensive detection of the currently difficult to call diseases-causing mutations in the complex gene families.

SPEAKER PROFILEDr. Drmanac is one of the founders of the field of Genomics. Currently he serves as Senior VP of BGI (Shenzhen, China) and Director and Chief Scientific Officer (CSO) at Complete Genomics, Inc. (CGI), that he co-founded with Cliff Reid in 2005 to develop technologies for efficient and accurate individual genome sequencing. CGI was acquired by BGI in 2013. Dr. Drmanac invented and led the development of massively parallel DNA sequencing using DNA nanoarrays, ligation-bases DNA sequencing and long fragment read technologies. Earlier, in 1994, he co-founded Hyseq (later Nuvelo and now ARCA), and served as Director until 2001. As SVP of research and CSO, he led Hyseq’s effort to discover and patent thousands of novel genes for its drug development pipeline. Prior to Hyseq, he was a group leader at Argonne National Labs (1991-1994) as part of DOE Human Genome Project. He completed his postdoctoral studies in 1990 at ICRF in London. He received his Ph.D. in Molecular Biology in 1988 at Belgrade University, Serbia, for the conception and pioneering development of the first massively parallel sequencing technology using DNA microarrays. At the same university he received BS (1981) and MS (1985) degrees in Molecular Biology. Since 1987, Dr. Drmanac and his teams received over 50 patents and over $20M in government grants and over $300M in investments for the development of advanced genome sequencing technologies.

Real World Data – Changing Drug Development Paradigm

ABSTRACTIn a broad sense, real world data (RWD) refer to data collected outside of traditional clinical trial settings. The availability and quality of RWD have been improving in recent years. The utility of RWD is also expanding and spans across research and development, regulatory approval and access, as well as medical practice. In the era of personalized medicine, the insights and evidence drawn from RWD have become even more relevant. With an increasing acceptance on using RWD to support decisions, how will the drug development paradigm change?

ABSTRACTAlzheimer’s disease and other dementing disorders pose important scientific biomedical and socio-economic problems that require a multi-pronged, interdisciplinary approach. In this lecture, Dr. Mucke will describe molecular and cellular processes thought to trigger or mediate Alzheimer’s disease, as well as synaptic and neural network dysfunctions that may underlie the inexorable cognitive decline and behavioral abnormalities it causes. He will also discuss how some of the latest mechanistic insights might be translated into better treatments for this illness through the development of novel therapeutics or the re-purposing of drugs used for other conditions.

SPEAKER PROFILEDr. Mucke is the founding director of the Gladstone Institute of Neurological Disease and holds joint appointments as the Joseph B. Martin Distinguished Professor of Neuroscience and Professor of Neurology at the University of California, San Francisco. He trained at the Georg-August University and the Max Planck Institute for Biophysical Chemistry in Gottingen, Germany, the Cleveland Clinic, the Massachusetts General Hospital and Harvard Medical School and The Scripps Research Institute.

Dr. Mucke’s research focuses on processes that result in memory loss and other major neurological deficits, with an emphasis on Alzheimer’s disease and related disorders. He has generated informative experimental models of these conditions and used them to identify disease mechanisms and novel strategies to prevent neurological decline.

For this contributions, Dr. Mucke has received the Potamkin Prize from the American Academy of Neurology, the MetLife Foundation Award for Medical Research, the Kalid Iqbal Lifetime Achievement Award and the Zenith Award form the Alzheimer’s Association, the American Pacesetter Award from the ARCS foundation, a MERIT award from the National Institute of Health, and an Award for Excellence in Direct Teaching and Mentoring from the Haile T. Development of New Therapeutic Agents Targeting Key

Cancer Stem Cell Pathways

ABSTRACTCancer stem cells (or tumor initiating cells) mediate tumor progression, metastasis, and recurrence after therapy. Using our platform of patient-derived xenografts, comprised of approximately 200 tumors, we have developed first-in-class biologic agents that block key CSC pathways including Notch, Wnt and RSPO-LGR. We have developed seven therapeutics currently in clinical testing - anti-DLL4 (demcizumab), anti-Notch2/3 (tarextumab), anti-Notch1 (brontictuzumab), anti-FZD (vantictumab), and Fzd8-Fc (ipacfricept) and anti-DLL4/VEGF. In addition, we have developed a new therapeutic agent, anti-Rspo3. These agents inhibit tumor growth through multiple mechanisms including a reduction of CSC frequency. The presentation will focus on the activity of anti-Rpso3 in colon, lung and ovarian cancers. We have found that anti-Rpso3 reduces tumor growth as a single agent and in combination with chemotherapeutic agents. Anti-Rspo3 treatment inhibits expression of stem cell markers and reduces tumorigenicity in serial transplantation studies. We have observed a strong correlation between Rpso3 expression and sensitivity to treatment, enabling a biomarker-focused strategy for patient selection and clinical development. Anti-Rspo3 (OMP-131R10) entered clinical testing in 2015.

SPEAKER PROFILEDr. Timothy Hoey is Senior Vice President, Cancer Biology at OncoMed Pharmaceuticals. The Cancer Biology group is responsible for testing new drugs targeting cancer stem cells and translational research to discover bio-markers indicative of CSC frequency and patients most likely to respond to OncoMed therapeutic antibodies. Dr. Hoey previously served as Director, Biology Department at Amgen (San Francisco) and was responsible for characterization of oncogenes and development of drugs to target oncogene products. Prior to this he was Director, Biology Department at Tularik, responsible for Cancer and Immunology research.

Prior to joining Tularik, he performed postdoctoral studies on the mechanisms of transcriptional regulation at U.C. Berkeley under Dr. Robert Tijian. Dr. Hoey received his Ph.D. from Columbia University, where he conducted research on regulation of gene expression and embryonic development with Dr. Michael Levine. Dr. Hoey is a co-inventor on several patents and has authored or co-authored over 60 scientific publications.

ABSTRACTResearch breakthroughs in the life science industry have opened doors for many exciting areas such as precision medicine, cancer immunotherapy, and mobile health. Intellectual property protection of new discoveries in these areas, however, has been more difficult to obtain than ever. During my talk, I will discuss some of the legal challenges a life science professional would be facing, pitfalls to avoid, and issues to consider in the course of obtaining valuable patent rights in these areas. I will also discuss unique considerations for patent rights arising from US/China collaborations.

SPEAKER PROFILEDr. Janet Xiao is a partner in Morrison & Foerster’s Life Science Group residing in the Palo Alto office. She primarily represents clients in the biotechnology and pharmaceutical industries in their world-wide patent procurement, patent portfolio management, and strategic planning. Dr. Xiao advises biopharmaceutical companies and research institutions on patent matters relating to various technologies including antibody therapeutics, nanomedicine, personalized medicine, drug delivery systems, drug screening platforms, diagnostics, and nutraceuticals.

Dr. Xiao also works extensively in performing IP due diligence reviews in the contexts of VC investments, technology transactions, mergers and acquisitions, as well as marketing and manufacturing clearances for biopharmaceutical products. She is among the very few IP attorneys who are both equipped with solid skills in global strategic IP management and knowledgeable about IP issue unique to China and Chinese clients. In both 2014 and 2015 Dr. Xiao was listed in Chambers Global as a leading IP expert in the U.S. and a leading IP lawyer in China as an expert based abroad.

“Organs, not Cells” – Generation of Functional Organs from iPS Cells via

Interspecies Blastocyst Complementation

ABSTRACTHematopoietic stem cells (HSCs) have played a flagship role in stem cell biology, providing a number of conceptual ideas and models. Among them is the concept of the “niche”, a special bone-marrow micro-environment that by exchanging cues regulates stem-cell fate (self-renewal, differentiation, hibernation, and proliferation). The concept of the stem-cell niche was further extended to generation of organs. We postulated that an “organ niche” exists in a developing animal and that this niche was empty when development of an organ is genetically disabled. This organ niche, we reasoned, should be compensated developmentally by blastocyst complementation using wild-type pluripotent stem cells. We provided proof of principle of organogenesis from xenogeneic pluripotent stem cells (PSCs). Demonstration of generation of a functional organ from PSCs in mice and pigs is a very important step toward generation of human cells, tissues, and organs from individual patients’ own PSCs in large animals.

SPEAKER PROFILEHiromitsu Nakauchi obtained a M.D. from Yokohama City University School of Medicine and a Ph.D. in immunology from University of Tokyo Graduate School of Medicine. He isolated CD8 genes during his post-doc period at the Laboratory of Prof. Leonard Herzenberg at Stanford University. After returning to Japan, he started working on hematopoietic stem cells in his laboratory at RIKEN. In 1994, he became Professor of Immunology at the University of Tsukuba where he demonstrated that a single hematopoietic stem cell could reconstitute the entire hematopoietic system, a definitive experimental proof for the “stemness”. Since April 2002, he has been a Professor of Stem Cell Therapy in the Institute of Medical Science at The University of Tokyo (IMSUT). In 2008, he was appointed Director of newly established Center for Stem Cell Biology and Regenerative Medicine at IMSUT. Just recently, he returned to Stanford University as a faculty to continue his stem cell research at the Institute of Stem Cell Biology and Regenerative Medicine. Goals of his work are to translate discoveries in basic research into practical medical applications.

1999 President’s MessageThe Chinese Bioscience Association started in late 90’s from a scatter of individual Chinese Bioscientists working at San Francisco Bay area. With few connections outside of our individual workplaces, many of our original members sought to socialize with other Chinese professionals in the bio-science field. Eventually, our casual socials turned increasing academic as our shared interests in bio-science compelled us to seek out more established individuals in the field to share their experiences and knowledge. With a desire to learn, therefore, our the humble group of Bio-scientists formed the beginning of CBA. Looking back now, with our fantastic annual conference and many members, it is amazing to see how much our organization has grown. As a past president, I am truly grateful that so many of you still share our passion for kinship and bioscience. I wish you all the best, and hope that you all enjoy another successful annual conference.
